چکیده

Incubation of adherent human breast epithelial HBL100 cells with epidermal growth factor (EGF) decreased [3H]dexamethasone binding by 35% with no effect on affinity. Maximal inhibition was obtained at 3 n\i EGF and the 50% effective dose was 0.2 mi EGF. Decreased dexamethasone binding induced by 3 n\i EGF was maximal by 5 min of treatment and, in the continuous presence of EGF, persisted at a constant level over 4 days. The action of EGF was antagonized by 12-O-tetradecanoylphorbol-13-acetate, which did not inhibit dexamethasone binding significantly, and by concanavalin A. In homogenates of EGF-treated cells, decreased dexamethasone binding was observed only in the cytosolic fraction. Saturation dexamethasone binding inhibited the growth rate of 11HI 100 cells by approximately 50%, but concurrent treatment with EGF over came this inhibition. The effect of EGF on dexamethasone-inhibited cell growth also was antagonized by 12-O-tetradecanoylphorbol-13-acetate.

Altered Expression of Epidermal Growth Factor Receptor (EGFR) in Glioma

EGFR is a key molecule in cancer cells. EGFR signaling was shown to promote tumor cell proliferation and survival, invasion and angiogenesis and mediate resistance to treatment, including ionizing radiation in preclinical models. We extracted proteins from astrocytoma (III and IV) oligodendroglioma(IV) tumors and normal brain tissues and then evaluated the protein purity by Bradford test ...

Ectopie Expression of Epidermal Growth Factor Receptors Induces Hormone Independence in ZR-75-1 Human Breast Cancer Cells1

Epidermal growth factor (EGF) receptor is inversely related to ex pression of estrogen receptor (ER) and progesterone receptor in primary breast tumors and is a negative predictor for response to endocrine therapy. To investigate a possible causal role of EGF receptor expres sion in breast cancer progression to hormone independence, we have created an experimental cell system. Epidermal growth ...

Triple tandem mimotope peptide of Epidermal Growth Factor Receptor displaying on the surface of M13 phage induces anti-tumor response in mice tumor model

Introduction: Epidermal growth factor receptor (EGFR) has been shown to play a critical role in tumor cell growth and its overexpression has been observed in many epithelial tumors. In the field of cancer vaccine research, displaying the peptide mimotope on the surface of phage particles has shown promising results. Methods: In this study using m13-PVIII phage display system, two constructs we...

2D-QSAR and docking studies of 4-anilinoquinazoline derivatives as epidermal growth factor receptor tyrosine kinase inhibitors

Introduction: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor derivatives play an important role in the treatment of cancer. We aim to construct 2D-QSAR models using various chemometrics using 4-anilinoquinazoline-containing EGFR TKIs. In addition, the binding profile of these compounds was evaluated using a docking study. Materials and Methods: In this study, 122 compounds of...
